San Marco is the beating heart of Venice. This is the sestiere (district) where all the big-hitters live: St Mark's Square, the Basilica, the Doge's Palace, the Bridge of Sighs, and the Rialto Bridge are all either inside San Marco or right on its borders. It's the most central part of the city, threaded with narrow calli (alleyways) that open onto grand piazzas, canal-side cafes, and some of Italy's most famous landmarks. Staying in San Marco puts you within walking distance of practically everything, and the vaporetto stops along the Grand Canal connect you to the rest of Venice in minutes.
